Target Information

USP7 (also known as HAUSP) is a ubiquitin-specific protease that removes ubiquitin from substrate proteins. This deubiquitylating activity stabilizes its substrates by preventing their proteasomal degradation. USP7 is known as a direct antagonist of Mdm2, the E3 ubiquitin ligase for tumor suppressor p53. Under normal conditions, Mdm2-mediated ubiquitylation keeps p53 levels low. In response to oncogenic stress, USP7 can deubiquitinate and stabilize p53, suggesting a tumor-suppressive role. Additionally, USP7 may participate in oncogenic stabilization of p53 through Myc or E1A pathways, potentially providing an alternative mechanism for cellular protection against oncogenic insults.
