Target Information

Major histocompatibility complex (MHC) class II molecules destined for presentation to CD4+ helper T cells are regulated by two key events:

  1. Dissociation of class II-associated invariant chain peptides (CLIP) from the antigen-binding groove in MHC II α/β dimers via MHC molecules HLA-DM and -DO.
  2. Subsequent peptide antigen binding.

HLA-DM and -DO molecules accumulate in endosomal/lysosomal compartments and on B cell surfaces, regulating CLIP dissociation and promoting exogenous peptide binding to HLA class II molecules (HLA-DR, -DQ, -DP).
RFLP analysis of HLA-DM genes from rheumatoid arthritis (RA) patients suggests certain polymorphisms may contribute to RA susceptibility.

HLA-B belongs to the HLA class I heavy chain paralogs, which play a central role in presenting peptides derived from the endoplasmic reticulum lumen. HLA-B and -C can form heterodimers with β2-Microglobulin, and polymorphisms yield hundreds of alleles.

HLA and MHC antibodies are essential in Immunopeptidomics, aiding in the identification and characterization of neoantigens via high-performance liquid chromatography coupled to tandem mass spectrometry.
