Product Specific Information
The UC10-4B9 monoclonal antibody reacts with mouse CD152, also known as cytotoxic T lymphocyte antigen-4 (CTLA-4). CTLA-4, structurally similar to CD28, is expressed at low levels on activated T cells and binds B7 family members CD80 (B7-1) and CD86 (B7-2) with higher affinity than CD28. While CD28 provides a costimulatory signal, CTLA-4 inhibits T cell activation by reducing IL-2 secretion and proliferation. A large portion of CTLA-4 is localized intracellularly due to ER retention motifs, thus intracellular staining may be necessary for complete detection.

Applications Tested:
Tested by intracellular staining and flow cytometric analysis of stimulated mouse splenocytes using the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00).

Target Information
CTLA4 (Cytotoxic T-Lymphocyte Antigen 4) is a CD28-family receptor mainly expressed on CD4⁺ T cells. It binds CD80 and CD86 on antigen-presenting cells with higher affinity than CD28, delivering inhibitory signals that suppress T cell activation. Intracellular CTLA4 is also found in regulatory T cells and contributes to their function.
