Product Specific Information

The 14D3 monoclonal antibody reacts with human CD152 (CTLA-4), a protein structurally similar to CD28. CTLA-4 is expressed on activated T cells and binds B7 family members CD80 (B7-1) and CD86 (B7-2) with higher affinity than CD28.
CTLA-4 delivers inhibitory signals to T cells, restricting activation by reducing IL-2 secretion and proliferation. The cytoplasmic domain contains ER retention motifs, leading to intracellular localization upon TCR signaling.
This antibody also recognizes rhesus monkey CTLA-4 and exhibits inhibitory activity.

Applications Tested: Pre-titrated and tested by intracellular staining and flow cytometry using the Intracellular Fixation and Permeabilization Buffer Set (Product # 88-8824-00) and BestProtocols® (Protocol A): Two-step protocol for cytoplasmic intracellular proteins.
Use at 5 µL (0.03 µg) per test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test.
For complete detection, assess both intracellular and surface CTLA-4 expression.

Target Information

CTLA4 (Cytotoxic T-Lymphocyte Antigen 4) is a CD28-family receptor expressed mainly on CD4+ T cells. It binds CD80 and CD86 with higher affinity than CD28, delivering inhibitory signals that prevent T cell activation. Intracellular CTLA4 is also found in regulatory T cells and contributes to their suppressive function.
