Target Information

Activation of NF-κB via Toll-like receptor (TLR) and IL-1 receptor signaling is a key component of innate immune responses. These signals are mediated by adaptor molecules such as TRIF, TIRAP, and MyD88. Regulatory mechanisms include inhibition of TLR expression/signaling by ST2 and SIGIRR, and ubiquitination of selected TLRs by TRIAD3A, an E3 ubiquitin-protein ligase.
TRIAD3A, a RING finger protein, binds TLR4 and TLR9 (and to a lesser extent TLR3 and TLR5), promoting ubiquitination and degradation of these receptors. Overexpression of TRIAD3A reduces TLR4/9 activation, while depletion enhances TLR signaling.
