Description:
Mouse IL-33 (also known as NF-HEV and DVS 27) is a 30 kDa proinflammatory cytokine produced by endothelial and epithelial cells. It is released during necrotic cell death and was first found in the nucleus of endothelial cells. This dual pattern of expression is characteristic of alarmins (endogenous danger signals) such as IL-1α and HMGB1. IL-33 serves as a ligand for ST2 (a member of the IL-1 receptor family), which is expressed on mast cells and Th2 effector T cells, contributing to Th2-mediated inflammation.
Although initially reported to be processed by Caspase-1, more recent data suggest that Caspase-3 or -7 process IL-33 at physiological concentrations. IL-33 induces IL-5 and IL-13 production, activating basophils, eosinophils, and mast cells. It may also modulate inflammation by interacting with IL-1. Mouse IL-33 shares 55% amino acid identity with human and 90% with rat IL-33.

IL-33 (Interleukin-33):
IL-33 is a 270 amino acid protein belonging to the IL-1 family with an IL-1-like C-terminal domain. It functions as both a proinflammatory cytokine and an intracellular nuclear factor with transcriptional regulatory properties. IL-33 binds to IL1RL1/ST2, recruiting MYD88, IRAK1, IRAK4, and TRAF6, leading to activation of NF-κB and MAP kinases and production of Th2-associated cytokines (IL-4, IL-5, IL-13).
In vivo, IL-33 induces Th2 cytokine expression and can cause eosinophilia, splenomegaly, and mucosal organ pathology. It is proteolytically converted to its mature form by CASP1 and is highly expressed in high endothelial venules (tonsils, Peyer’s patches, mesenteric lymph nodes), but nearly absent in placenta. IL-33 may co-localize with heterochromatin and act as a transcriptional repressor, supporting its dual role as both cytokine and nuclear factor.
