Description:
The NOK-1 monoclonal antibody reacts with human Fas (CD95) Ligand, a 40 kDa type II transmembrane glycoprotein. FasL is a member of the TNF family expressed by neutrophils, monocytes, activated T cells, and NK cells. Its interaction with Fas receptor induces Fas-mediated lymphocyte killing. FasL can be cleaved by MMPs to form a soluble 26 kDa protein; thus, MMP inhibitors are recommended for optimal surface detection.

• Blocking of FasL-mediated killing in functional assays

Applications Tested:
Validated for flow cytometric analysis of normal human peripheral blood cells. Recommended ≤1 µg per test (100 µL final volume). Cell number can range from 10⁵–10⁸ cells/test. Antibody titration is recommended for optimal performance.

Target Information

CD178 (Fas ligand, FasL) is a type-II membrane protein with its N-terminus in the cytoplasm and C-terminus extracellular. Its receptor, FasR, is a type-I membrane protein of the TNF/NGF receptor family. CD178 binding to FasR triggers apoptosis via receptor oligomerization.

CD178 exists in membrane-bound (45 kDa) and soluble (26 kDa) forms, cleaved by metalloproteinases. It is expressed in activated T lymphocytes, NK cells, and immune-privileged tissues (testis, eye), as well as thymus, liver, ovary, lung, heart, and kidney. CD178-mediated apoptosis is involved in antiviral immune responses and immune regulation. Dysregulation has been linked to atherosclerosis, hypertension, AIDS, hepatitis, and cancer.
