Description:
The HT52 monoclonal antibody reacts with human Toll-like receptor (TLR) 4. The Toll family includes at least ten members in humans, characterized by an extracellular domain with leucine-rich repeats and a cytoplasmic domain similar to the type I IL-1 receptor.

TLR2 and TLR4 are pattern recognition receptors involved in innate immunity and inflammation. TLR4 associates with MD-2 and CD14 to form a complex responsible for LPS recognition and signaling. TLR4 is expressed by peripheral blood monocytes, with variable expression among individuals.

The HT52 antibody exhibits neutralizing activity and is suitable for functional assays and flow cytometric analysis. Tested by flow cytometry on TLR4-transfected cells, it can be used at ≤0.25 µg/test (100 µL final volume). Optimal concentration should be titrated per assay.

Target Information

TLR4 (Toll-like receptor 4) is a key component in pathogen recognition and activation of innate immunity. It is most abundantly expressed in placenta and myelomonocytic leukocytes. TLR4 forms part of the LPS receptor complex with CD14 and LY96, mediating responses to gram-negative bacterial lipopolysaccharides (LPS).

Mutations in TLR4 can affect LPS responsiveness, and multiple transcript variants exist. Expression is observed in monocytes, a subset of B-cells, and placenta. Studies in TLR4-deficient mice confirm its essential role in LPS recognition and resistance to Gram-negative sepsis.
