Product Specific Information
The HTA125 monoclonal antibody reacts with human Toll-like receptor 4 (TLR4). TLR4 is a type I transmembrane protein involved in innate immunity and inflammation. It forms a complex with MD-2 and CD14 for LPS recognition and signaling. TLR4 is expressed on peripheral blood monocytes, with variable expression among donors.
For optimal staining of TLR4, it is recommended to use the lysed whole blood protocol rather than Ficoll-gradient prepared cells, as density gradient preparation can reduce staining intensity.

Recommended Protocol:
For detecting peripheral monocytes, use a three-step staining protocol:
- Purified anti-human TLR4
- Biotin anti-mouse IgG
- Streptavidin-PE
Applications Tested:
Tested by flow cytometric analysis of normal human peripheral blood cells. Use ≤1 µg per test (100 µL final volume, 10⁵–10⁸ cells/test). Titrate for optimal performance.
Target Information
TLR4 (Toll-like receptor 4) is a key component of the innate immune system, recognizing pathogen-associated molecular patterns (PAMPs) and mediating cytokine production. It is most abundantly expressed in placenta and myelomonocytic leukocytes. TLR4 forms part of the LPS receptor complex with CD14 and LY96, initiating signaling in response to Gram-negative bacterial lipopolysaccharides.
Mutations in TLR4 affect LPS responsiveness. Several transcript variants exist, though their protein-coding potential is uncertain. TLR4 expression is observed in monocytes, a subset of B cells, and placental tissue. TLR4-deficient mice demonstrate increased susceptibility to Gram-negative sepsis, confirming its role in LPS recognition.
