This RR3-15 monoclonal antibody reacts with the mouse T-cell receptor (TCR) V beta 11 chain.
TCR specificity is determined by Va, Ja, Vb, Db, and Jb gene rearrangement.
The RR3-15 antibody recognizes the V beta 11 chain on T cells from mouse strains with the b haplotype (e.g., C57BL/6, B10), but not in mice with the a haplotype (e.g., C57L, SJL, SWR).

• Vbeta11+ T cells are eliminated in mice expressing MHC Class II I-E molecule.
• V beta 11+ CD4+ T cells have been associated with graft-versus-host disease (GVHD) severity.
• RR3-15 antibody may activate V beta 11+ T cells.

Applications Tested: Tested by flow cytometry on mouse lymph node cells.
Recommended ≤0.125 µg/test (100 µL final volume; 10⁵–10⁸ cells/test).
Titrate antibody for optimal performance.

Fluorophore Information:
PerCP-eFluor® 710 emits at 710 nm and is excited with the blue laser (488 nm).
Can replace PerCP-Cyanine5.5.
Recommended filter: 710/50 bandpass (695/40 acceptable alternative).
Ensure instrument compatibility.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation.
Protect vial and stained samples from light.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using:

• IC Fixation Buffer (Product #00-8222)
• 1-step Fix/Lyse Solution (Product #00-5333)

Performance after fixation may vary by clone and should be validated empirically.

Recognition of self-peptides is linked to autoimmune diseases, while mutant self-peptides relate to tumors.
Superantigen engagement of TCRs can lead to toxic shock syndrome.
Autoantibodies to TCR V beta segments have been found in rheumatoid arthritis and systemic lupus erythematosus, potentially modulating immune responses.
Most human T cells express TCR alpha-beta with CD4 or CD8, while a subset of double-negative (DN) T cells is associated with autoimmune and immunodeficiency disorders.
Gamma-delta T cells, found in epithelial tissues, show limited TCR diversity and distinct antigen recognition, with roles in antitumor and immunoregulatory activity.
