Product Specific Information

Description:
The SH2.1 monoclonal antibody reacts with mouse Siglec-G (sialic acid-binding immunoglobulin-like lectin-G). Siglec-G is an inhibitory receptor expressed specifically in B cells, with B1 B cells showing the highest expression levels. Overexpression of Siglec-G inhibits B cell receptor-mediated calcium signaling. Siglec-G-deficient mice display expansion of the B1a cell population and increased natural IgM antibody titers. These cells exhibit decreased spontaneous apoptosis and prolonged lifespan, potentially due to elevated NFATc1 expression. Siglec-G-dependent negative regulation explains the muted signaling response of B1 cells.

Applications Reported:
This SH2.1 antibody has been reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometry using mouse splenocytes. Recommended use is ≤ 0.125 µg per test (in 100 µL final volume). Optimal titration should be empirically determined.

Fluorophore Information:
PerCP-eFluor® 710 emits at 710 nm and is excited with the 488 nm blue laser. It can substitute for PerCP-Cyanine5.5. Recommended filter: 710/50 bandpass (695/40 acceptable alternative). Ensure instrument compatibility.

Fixation:
Samples may be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark. Clone-specific performance should be empirically verified.

Target Information

Siglec-G is an inhibitory receptor expressed in B cells, particularly B1 cells. Overexpression inhibits B cell receptor-mediated calcium signaling. Siglec-G-deficient mice show B1a cell expansion, elevated natural IgM titers, reduced apoptosis, and prolonged lifespan due to increased NFATc1 expression. This receptor mediates negative regulation in B1 cells, contributing to their muted signaling response.
