The 6B11 monoclonal antibody recognizes an epitope of the CDR3 region formed by the germ-line configuration of Vα24 and Jα18 of the TCR α locus, resulting in expression of an invariant T-cell receptor (TCR) specific to invariant NK T cells (iNKT). This receptor is homologous to murine Vα14 Jα18 and pairs with restricted TCR β chains, predominantly Vβ11 in humans.

iNKT cells are CD1d-restricted and respond to glycolipid antigens presented by CD1d, rapidly producing IL-4, IL-13, and IFN-γ. Despite NK-like cytolytic activity, iNKT cells are key regulators of immune responses through rapid cytokine production in autoimmune, infectious, and cancer contexts. The proportion of iNKT cells in normal human peripheral blood varies between 0.01–1% of total T cells.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and validated for human peripheral blood cells at 5 µL (0.25 µg) per test (100 µL final volume). Recommended cell count: 10⁵–10⁸ cells/test.

PerCP-eFluor® 710 can substitute for PE-Cy5, PE-Cy5.5, or PerCP-Cy5.5. It emits at 710 nm and is excited by the 488 nm blue laser. Recommended filter: 685 LP dichroic mirror and 710/40 band pass filter (695/40 acceptable).

Target Information

T cell receptors (TCRs) enable T cells to distinguish self from foreign peptides presented by MHC class II molecules, a critical function for adaptive immunity. Aberrant TCR recognition of self-peptides is linked to autoimmune diseases, while mutated self-peptides are associated with tumors.

Superantigen-mediated TCR engagement can lead to toxic shock syndrome. Autoantibodies targeting TCR Vβ segments have been identified in rheumatoid arthritis (RA) and systemic lupus erythematosus (SLE), potentially modulating inflammatory responses.

Most human T cells express TCR αβ with CD4 or CD8 (single positive), though a small subset lacks both (double negative). Increased αβ double-negative T cells are observed in certain autoimmune and immunodeficiency disorders. γδ T cells, primarily epithelial, exhibit limited TCR diversity and distinct antigen recognition, with subsets showing antitumor and immunoregulatory functions.
