Product Specific Information

The eBioOMAK-D antibody reacts with mouse perforin (pore-forming protein, pfp, Prf). Perforin is a cytolytic mediator found in cytotoxic T lymphocytes (CTLs) and natural killer (NK) cells, playing a crucial role in immune responses against tumors and viral infections.

By immunoblotting, eBioOMAK-D recognizes a ~70 kDa band in lysates of CTLL-2 mouse cytotoxic cells and IL-2 stimulated splenocytes, but not in unstimulated ones. Flow cytometric analysis shows increased staining upon IL-2 or anti-CD3/CD28 stimulation. Perforin knockout splenocytes do not stain or show detectable protein by western blotting.

In IL-2 stimulated splenocytes:

• NK cells (CD49b+) contain perforin.
• CD8 cells show variable expression depending on culture conditions.
• Anti-CD3/CD28 stimulation increases perforin expression in both NK and CD8 cells.

eBioOMAK-D is also cross-reactive to human perforin and co-stains CD56+ PBMC cells.

Granzyme B and perforin expression may not correlate; Granzyme B increases earlier and more dramatically (>10,000-fold mRNA increase) compared to perforin (10–100-fold).

For intracellular staining and flow cytometry, use the Foxp3 buffer system (Product #00-5523) for optimal results.

Perforin is a major cytolytic protein stored in cytoplasmic granules of cytotoxic lymphocytes. It plays an essential role in immune defense against tumors and viruses.

• Length: 555 amino acids (includes 21-aa signal peptide)
• Mechanism: Pore-forming, similar to complement component C9
• Expression: Found in killer cell lines, absent in helper T cells and other tumor cells
• Clinical Relevance: Mutations cause familial hemophagocytic lymphohistiocytosis type 2 (HPLH2), a rare autosomal recessive disorder.
• Alternative splicing produces multiple transcript variants.
