Product Specific Information

The MC10 monoclonal antibody recognizes all forms of human estrogen receptor (ER) beta. ER beta is a member of the nuclear receptor superfamily, functioning as a ligand-dependent transcription factor. Upon estrogen binding, ER beta dissociates from chaperone proteins and dimerizes with ER beta or ER alpha. Unlike ER alpha, ER beta signaling is considered anti-proliferative and pro-apoptotic.
MC10 clone detects full-length ER beta and all known splice variants.

Applications Reported:
Immunoprecipitation, Western blotting, immunohistochemical staining of formalin-fixed paraffin-embedded tissue sections, microscopy, ELISA, and immunocytochemistry.

Applications Tested:
Validated for IHC on formalin-fixed paraffin-embedded human tissue using low or high pH antigen retrieval at ≤30 µg/mL. Antibody titration is recommended for optimal performance.


Target Information

Estrogen Receptor beta (ERβ, NR3A2) is approximately 55 kDa, with isoforms ranging from 36–59 kDa. Initially cloned from human testis, ERβ belongs to the steroid/thyroid hormone receptor superfamily. Upon estrogen binding, ERβ undergoes conformational change, dimerizes, and binds to estrogen response elements (EREs) in DNA, regulating transcription via coactivator or corepressor recruitment.
Two receptor forms exist, ERα (ESR1) and ERβ (ESR2), both with similar estrogen-binding affinities. Certain ERβ isoforms inhibit ERα activity in reproductive tissues. ERβ is broadly expressed and acts as a tumor suppressor, implicated in prostate cancer, autism spectrum disorder, medullary thyroid carcinoma, De Quervain disease, and cardiovascular disease risk.
