For direct ELISA, coat wells with 1% glutaraldehyde for 5 minutes at room temperature, followed by 3×PBS wash steps prior to coating with hyaluronic acid.
Sheep anti-hyaluronic acid antibody recognizes hyaluronic acid, a mucopolysaccharide that is an integral part of the gel-like substance of animal connective tissue, serving as a lubricating and protective agent.

Target Information

Hyaluronic acid (HA) is the most abundant glycosaminoglycan (GAG) in mammalian tissue. It is present in high concentrations in connective tissues such as skin, vitreous humor, cartilage, and umbilical cord, with the largest reservoir in synovial fluid (SF) of diarthrodial joints (0.5–4 mg/mL).
HA is the major hydrodynamic nonprotein component of joint SF, conferring viscoelastic, shock-absorbing, and lubricating properties. Its macromolecular size and hydrophilicity help retain fluid in the joint cavity while regulating solute exchange between synovial capillaries and cartilage.
HA interacts with cell receptors such as CD44 and RHAMM, modulating proliferation, migration, and gene expression. Its biological properties are molecular weight-dependent, arising from its unique unbranched polysaccharide structure composed of repeating disaccharides of N-acetylglucosamine and glucuronic acid.
Within the joint cavity, HA is primarily synthesized by type B synovial cells.
