MA1-164 detects glycosylated LAMP-1 at approximately 130 kDa.
M-PER (Product #78501), IP Lysis Buffer (Product #87787), or a 1% Triton X-100 lysis buffer are recommended for Western blot.
LAMP-1 was not detected when RIPA buffer was used to generate cell lysates.

Target Information

LAMP1 (CD107a, lysosome-associated membrane protein-1) together with LAMP-2 is a major constituent of lysosomal membranes.
Approximately 1–2% of total CD107a is also found on the plasma membrane.
LAMP1 is a heavily glycosylated membrane protein containing:

• A signal peptide
• 18 sites for N-linked glycosylation
• A single membrane-spanning segment
• A short cytosolic tail (11 amino acids)

LAMP proteins are involved in lysosome biogenesis and required for lysosome-phagosome fusion.
LAMP1 is a type I integral membrane protein transported from trans-Golgi network to endosomes and lysosomes.
Upon cell activation, LAMP1 translocates to the plasma membrane via a carboxyl-terminal tyrosine-based motif (YXXI).
Perturbation of this motif disrupts lysosomal localization, causing cycling between plasma membrane and endosome.

Cell surface LAMP1 (and LAMP2) promote adhesion of human PBMCs to vascular endothelium, suggesting involvement in inflammatory responses.
Increased LAMP1 immunoreactivity is observed in neurons and glial cells surrounding senile plaques in Alzheimer’s disease and in medullary epithelial cells, macrophages, and lymphocytes during acute thymic involution.
LAMP1 is a reliable marker of mast cell activation.
