Target Information

The PAX genes are a family of transcription factors active in specific tissues during early embryonic development. Pax family members possess a DNA-binding domain encoded by the paired box, enabling them to regulate genetic programs.
Several Pax genes are associated with developmental mutations:

  • PAX3: Waardenburg syndrome
  • PAX6: Aniridia
  • PAX2: Wilms tumor

During embryogenesis, PAX2 is expressed in the developing kidney, particularly in condensing metanephric mesenchyme and early epithelial structures derived from mesenchyme. PAX2 mRNA and protein levels decrease as the tubular epithelium matures. However, PAX2 expression persists in the undifferentiated epithelium of Wilms tumors, correlating with poorly differentiated epithelial cell proliferation.
The Wilms tumor suppressor protein WT1 coincides with the down-regulation of PAX2, suggesting WT1 directly represses PAX2 transcription.
