Product Specific Information
This antibody specifically recognizes the cytoplasmic domain of the c-Met protein.
On Western blot, it detects a band at approximately 145 kDa. Reactivity was confirmed with human HT-29 breast carcinoma lysates and recombinant c-Met cytoplasmic domain protein.
It also shows reactivity in formalin-fixed, paraffin-embedded breast carcinoma tissues by immunohistochemistry.
For IHC using formalin-fixed, paraffin-embedded tissues, heat-induced epitope retrieval (HIER) with citrate buffer (pH 6.0) is required prior to staining.
Recommended antibody incubation period: overnight at 4°C.
Target Information
MET (c-Met) is a receptor-like tyrosine kinase associated with numerous human malignancies.
Upon ligand activation, MET interacts with PI3-kinase subunit PIK3R1, PLCG1, SRC, GRB2, and STAT3, leading to activation of signaling pathways such as RAS-ERK, PI3K-AKT, and PLCγ-PKC.
It plays key roles in embryonic development (gastrulation, muscle and neuron formation, angiogenesis, kidney development) and in adults contributes to wound healing, organ regeneration, and tissue remodeling.
MET dysregulation is linked to cancers including gastric, renal, and breast, making it a target for cancer therapeutics and diagnostics.
