이 항체는 약 22–23 kDa 크기의 Claudin-2 단백질에 특이적으로 반응합니다.
인간, 마우스, 랫, 견의 간 및 신장 조직, MDCK 및 Caco-2 세포 라이세이트에서 Western blot과 면역형광법으로 반응성이 확인되었습니다.
FFPE 조직(인간 정상 대장, 비장, 갑상선, 대장 및 유방암 조직)에서도 면역조직화학법으로 반응이 검증되었습니다.
- FFPE 조직의 IHC에서는 1–2 µg/mL 농도 사용 권장
- 염기성 EDTA buffer(pH 8.0)를 이용한 HIER 전처리 필요
- 일부 라이세이트에서 27 kDa의 비특이적 밴드가 관찰될 수 있음
- Caco-2 세포에서는 비특이적 밴드가 22 kDa 밴드보다 강하게 나타날 수 있음

Target Information
Tight junction은 상피세포 간의 특수한 접합 구조로, 세포 간 물질 이동을 차단하고 세포 극성을 유지하는 데 중요한 역할을 합니다.
Claudin-1과 Claudin-2는 이러한 tight junction의 주요 구조적 구성요소이며, Occludin과 달리 네 개의 막관통 영역을 가지는 약 22 kDa 단백질입니다.
Claudin-2는 ZO-1을 통해 actin 세포골격과 연결되며, 양이온(Na⁺) 선택성을 가지는 paracellular 채널로 작용합니다.
주로 간과 신장에서 발현되며, 소량은 뇌에서도 발견됩니다.
