Target Information

Epithelial sodium channels are amiloride-sensitive members of the Degenerin/epithelial sodium channel (Deg/ENaC) superfamily of ion channels. Members share structural similarity with two short intracellular termini, two membrane-spanning segments, and a large extracellular loop containing a conserved cysteine-rich region.
ENaC isoforms (α, β, γ) play essential roles in sodium and fluid balance in kidney, lung, and colon. They also mediate aldosterone-dependent sodium reabsorption in the distal nephron, regulating blood pressure.
ENaC is partly regulated through association with CFTR chloride ion channels. Gain-of-function mutations in β or γ ENaC can cause severe arterial hypertension (Liddle’s syndrome), while loss-of-function mutations in α or β ENaC lead to pseudohypoaldosteronism (PHA-1).
