Product Specific Information

The HTA125 monoclonal antibody reacts with human Toll-like receptor 4 (TLR4). TLR4 is a type I transmembrane protein involved in innate immunity and inflammation, forming a complex with MD-2 and CD14 for LPS recognition and signaling. It is expressed primarily in peripheral blood monocytes.

HTA125 immunoprecipitates human TLR4 (~100 kDa) from transfected cells. TLR surface expression varies among donors and is generally low on monocytes and other cell types.

Recommended Protocol:
For optimal staining, use lysed whole blood rather than Ficoll-gradient prepared samples, as density gradient preparation reduces staining intensity.

Applications Reported:
Validated for flow cytometric analysis.

Applications Tested:
Pre-diluted and tested at 5 µL (0.5 µg)/test for flow cytometry using human TLR4-transfected cells.
A test corresponds to staining a 100 µL cell sample (10⁵–10⁸ cells/test).

Fluorochrome Information:
Super Bright 600 is a tandem dye excited by the violet laser (405 nm) and emits at 600 nm. Use a 610/20 bandpass filter.
For multiple Super Bright dyes, use Super Bright Complete Staining Buffer (Product # SB-4401-75) to minimize non-specific interactions.

Light Sensitivity:
Protect vial and stained samples from light to prevent photo-induced oxidation.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-57) with minimal impact on brightness.

Excitation / Emission / Laser:
405 nm excitation, 600 nm emission, Violet laser.

Target Information

TLR4 is a member of the Toll-like receptor family that recognizes pathogens and activates innate immunity. It is expressed in placenta and myelomonocytic leukocytes. TLR4 forms part of the LPS receptor complex (CD14, LY96, TLR4), mediating cytokine production for immune response. Mutations in TLR4 affect LPS responsiveness. Expression occurs in monocytes, B-cells, and placenta. TLR4-deficient mice show increased susceptibility to Gram-negative sepsis.
