Product Specific Information

The ISA-3 monoclonal antibody reacts with human ICOS (Inducible COStimulatory molecule), also known as H4, CRP-1 and AILIM. ICOS is a T cell-specific activation molecule and a third member of the CD28/CTLA-4 family. It plays a key role in T cell activation and humoral immune responses, particularly in memory B cell and plasma cell generation.

Plate-bound ISA-3 induces production of IL-4, IL-5, IL-10, and other cytokines, but not IL-2. ISA-3 was generated against human ICOS antigen.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells. Recommended 5 µL (0.125 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Super Bright 600 is a tandem dye excited by the violet laser (405 nm) and emits at 600 nm. Recommended detection filter: 610/20 bandpass.

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity: Protect vial and stained samples from light.
Fixation: Samples may be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333).

ICOS (CD278) encodes a protein belonging to the CD28 and CTLA-4 receptor family. It functions as a homodimer and is involved in cell-cell signaling, immune response regulation, and cell proliferation. ICOS is associated with disorders such as Immunodeficiency, Common Variable, 1 and Common Variable Immunodeficiency.
