Product Specific Information

The YGIC86 monoclonal antibody reacts with mouse Glycoprotein A Repetitions Predominant (GARP, also known as Lrrc32 or Garpin). GARP is an ~80 kDa glycoprotein expressed on the cell surface. Expression has been reported in placenta, lung, kidney, heart, ovary, liver, skeletal muscle, and pancreas, and on megakaryocytes, platelets, and activated regulatory T (Treg) cells. GARP expression on Treg cells is essential for suppressive function, possibly as a receptor for LAP/TGF beta.

When analyzing platelet GARP expression, use the Foxp3 Staining Buffer Set (Product # 00-5523) for fixation before staining. Fixation/permeabilization is not required for Treg cells.
YGIC86 does not cross-react with rat GARP.

Applications Tested: Tested on stimulated mouse spleen cells at ≤0.125 µg/test (100 µL final volume). Optimize titration for best performance.

Super Bright 600 is a tandem dye excited by a 405 nm violet laser and emits at 600 nm (recommended filter: 610/20 bandpass). Ensure instrument compatibility.

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific interactions.

Light Sensitivity: Protect from light to avoid photo-induced oxidation.
Fixation: Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) with minimal impact on brightness or FRET efficiency.

Target Information

GARP (leucine rich repeat containing 32) is a type I membrane protein with 22 leucine-rich repeats and a short intracytoplasmic region. Identified via gene expression profiling of Treg and Th cells after TCR stimulation, GARP is expressed in CD4+, CD25+, FoxP3+ Treg cells and may serve as a surface marker for Treg identification.
