Product Specific Information

The monoclonal antibody TB28-2 recognizes the kappa light chain of human immunoglobulin (~22 kDa), composed of one variable and one constant region. Two kappa chains with heavy chains form the complete immunoglobulin structure. Expression of Ig kappa is typically found on more than half of B lymphocytes.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells.
Use at 5 µL (0.5 µg) per test (100 µL final volume). Cell number may range from 10⁵–10⁸ cells/test.

Fluorophore Information:
Super Bright 600 is a tandem dye excited by the violet laser line (405 nm) and emits at 600 nm. Recommended filter: 610/20 bandpass. Ensure instrument compatibility.

Staining Recommendation:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity:
Sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-57) for up to 3 days in the dark at 4°C with minimal impact on brightness and FRET efficiency.

Target Information

Immunoglobulin classes share a basic four-chain structure: two heavy chains and two light chains (kappa, lambda; ~22.5 kDa each). Kappa and lambda differ in their constant region antigenicity, with a typical ratio of 70:30.
Antibodies to kappa light chain are useful for identifying leukemias, plasmacytomas, and certain non-Hodgkin’s lymphomas. Demonstration of clonality in lymphoid infiltrates indicates malignancy.
