Product Specific Information
The monoclonal antibody XLP-1D12 reacts with human SLAM-associated protein (SAP, also known as SH2D1A). SAP is a 15-kDa SH2 domain-containing adaptor protein. Upon CD150/SLAM receptor ligation, SAP recruits Fyn tyrosine kinase to activate downstream signaling. It is expressed in thymocytes, T cells, NK cells, and NKT cells.
SAP plays roles in follicular T helper cell differentiation, NKT cell development, Th1/Th2 differentiation, and regulation of NK and CD8+ T cell cytotoxicity. Mutations in SAP cause X-linked lymphoproliferative (XLP) syndrome, associated with impaired humoral immunity and susceptibility to Epstein-Barr virus and lymphoma. SAP is also linked to autoimmune diseases such as systemic lupus erythematosus.
Applications Reported: Suitable for intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and validated for intracellular staining using the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00) and BestProtocols® Protocol B.
Use at 5 µL (0.5 µg) per test (100 µL final volume). Typical cell range: 10⁵–10⁸ cells/test.

Target Information
This gene encodes a protein involved in bidirectional stimulation of T and B cells. It contains an SH2 domain and associates with signaling lymphocyte-activation molecules, modulating SHP-2 recruitment and signal transduction. Mutations cause X-linked lymphoproliferative syndrome (Duncan disease), characterized by susceptibility to Epstein-Barr virus infection, severe mononucleosis, and malignant lymphoma. Multiple transcript variants exist.
