Description:
This MABA223 monoclonal antibody recognizes human E4BP4 (NFIL3), a basic leucine zipper transcription factor involved in transcriptional control of the mammalian circadian clock, motor neuron growth, von Willebrand factor expression, and osteoblast function. It is essential for NK, NKT, and CD8α⁺ dendritic cell development. Although high mRNA levels are seen in resting NK/NKT cells, E4BP4 protein expression is detected only after activation.

Applications Reported:
Intracellular staining followed by flow cytometric analysis.

Applications Tested:
Pre-titrated and validated for intracellular staining and flow cytometric analysis using stimulated human peripheral blood cells with the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00).
Use 5 µL (0.125 µg) per test in a 100 µL staining volume. Suitable for 10⁵–10⁸ cells/test.

Excitation/Emission:
488–561 nm excitation; 578 nm emission. Compatible with Blue, Green, and Yellow-Green lasers.

Filtration:
0.2 µm post-manufacturing filtered.

Target Information

Expression of interleukin-3 (IL3; MIM 147740) is restricted to activated T cells, NK cells, and mast cell lines. Transcription initiation depends on NFIL3, which binds to regulatory regions upstream of the transcription start site (Zhang et al., 1995 [PubMed 7565758]).
