Target Information

CXCR4 (also known as Fusin, LESTR, HUMSTR) is a member of the G-protein-coupled chemokine receptor family with seven membrane-spanning domains. It functions as a co-receptor for X4 HIV-1 entry into CD4+ cells and also serves as a co-receptor for HIV-2 binding through envelope glycoprotein gp120. CXCR4 mediates signal transduction via CXCL12/SDF-1 binding, leading to increased intracellular calcium and MAPK1/MAPK3 activation.

CXCR4 plays essential roles in:

  • Hematopoiesis and cardiac ventricular septum formation
  • Gastrointestinal vascularization via endothelial remodeling
  • Neuronal survival in the hippocampus and cerebellar development
  • Mediating LPS-induced inflammatory responses, including TNF secretion

Antibodies to CXCR4 can block HIV-1 and HIV-2 fusion and infection in human target cells. The amino-terminal domain and second extracellular loop of CXCR4 serve as HIV binding sites.
