The SMARTer Mouse TCR a/b Profiling Kit provides a powerful solution for T-cell receptor (TCR) repertoire analysis using next-generation sequencing (NGS). It employs a 5’-RACE-based approach to capture complete V(D)J variable regions of TCR transcripts from as little as 10–500 ng of total RNA (from mouse spleen, thymus, or PBMCs) or 1,000–10,000 purified T cells.
This kit enables the generation of data for both TCR-alpha and TCR-beta chain diversity, either in the same experiment or separately.

Technology Overview

Unlike multiplex PCR-based TCR profiling, this kit uses a single primer pair for each TCR subunit during library amplification, minimizing primer-dimer formation and sequence bias. The ~4-hour protocol yields indexed libraries ready for sequencing on Illumina platforms.
The kit leverages Takara’s proven SMART (Switching Mechanism at 5′ End of RNA Template) technology combined with LNA technology for unbiased and sensitive amplification of TCR mRNA sequences.
