Product Specific Information

The CX5 monoclonal antibody reacts with mouse NKG2D, a lectin-like molecule expressed on NK cells. Mouse NKG2D binds to RAE-1 (α, β, γ, δ, ε) and H60 molecules, co-stimulating NK activation receptors via DAP12/DAP10 adaptors.

Applications Reported:

• Flow cytometric analysis
• Blocking of NKG2D-ligand binding (RAE-1, H60)
• Inhibition of NK cell-mediated cytotoxicity against ligand-bearing tumors (in vitro, in vivo)

Applications Tested:

• Tested by flow cytometric analysis of mouse splenocytes
• Recommended ≤ 0.5 µg per test (100 µL final volume, 10⁵–10⁸ cells/test)
• Use Biotin anti-rat secondary antibody (Product #12-5783-81)
• Expression confirmed in C57BL/6 and NWNA mice; not detected in Balb/c or DBA/2
• No cross-reactivity to Ly-49A, C, D, or G2 observed
• 3D10 antibody induces lysis of target cells in culture

Additional Notes:
Each NovaFluor conjugate or kit is shipped with CellBlox Blocking Buffer. Add 5 µL per sample when staining with NovaFluor conjugates.
For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

The NKG2 gene family encodes type II transmembrane proteins with C-type lectin domains, located within the NK gene complex.
