Product Specific Information
The MD5-1 monoclonal antibody reacts with mouse DR5 (TRAIL-R2, Apo2, TRICK2, KILLER). DR5 binds to TRAIL, activates NF-kappaB, and induces TRAIL-mediated apoptosis. DR5 mRNA is broadly expressed in normal tissue, and human antigen is also expressed by some tumor cell lines.

Applications Tested:
The MD5-1 antibody has been tested by flow cytometric analysis of mouse DR5-transfected cells. It can be used at ≤0.25 µg per test (100 µL final volume). Cell number should be determined empirically (10⁵–10⁸ cells/test). Antibody titration is recommended for optimal assay performance.

Target Information
DR5 (Apo2, TRAIL-R2, TRICK2, KILLER) is a death domain-containing receptor for TRAIL, mediating apoptosis. It belongs to the TNF-receptor superfamily and can be activated by TNFSF10/TRAIL/APO-2L. FADD adaptor protein is required for DR5-mediated apoptosis. Two transcript variants and one non-coding transcript exist. Overexpression induces apoptosis and activates NF-kappa-B. DR5 is highly expressed in lymphocytes and spleen. Dysfunction is associated with squamous cell carcinoma and diffuse infiltrative lymphocytosis syndrome.
