Description:
The MIH18 monoclonal antibody reacts with human B7-DC (PD-L2), a member of the B7 family with an approximate molecular weight of 25 kDa. It is primarily expressed by dendritic cells and monocytes/macrophages. Unlike other B7 family members, B7-DC does not bind CD28/CTLA-4 but serves as a ligand for PD-1. MIH18 functions as a blocking antibody and is a valuable tool for studying PD-L2’s role in APC/T cell interactions and expression patterns in humans.

Applications Tested:
Tested on mature dendritic cells and normal human peripheral blood cells. Use ≤0.5 µg per test (100 µL final volume). Cell number per test may vary from 10⁵ to 10⁸ cells and should be optimized empirically.

Programmed death-ligand 2 (PD-L2/B7-DC):
PD-L2 is a member of the B7 ligand family within the immunoglobulin superfamily and acts as a ligand for PD-1. It is mainly expressed in dendritic cells but can be induced in various immune and non-immune cells depending on cytokine conditions (IL-4, TNF-α, IFN-γ). PD-L2 has 2–6 times higher affinity for PD-1 compared to PD-L1. It plays a dual role in immune regulation—both inhibitory and stimulatory—depending on receptor interactions. PD-L2 also binds to RGMb, influencing respiratory tolerance via BMP/neogenin signaling.
The human PD-L2 monomer consists of a 201-aa extracellular domain, a 21-aa transmembrane domain, and a 32-aa cytoplasmic domain.
