Product Specific Information

This 15-414 monoclonal antibody reacts with human CD172a (SIRP alpha), a cell surface glycoprotein with three Ig-like extracellular domains and two cytoplasmic ITIM motifs. These motifs recruit SHP-1 and SHP-2 phosphatases. CD172a is expressed on monocytes, granulocytes, dendritic cells, and CD34⁺CD38⁻ progenitor cells, but not on T or B lymphocytes. Neurons also express CD172a. Its ligand, CD47, mediates dendritic cell–T cell activation, neutrophil migration, and phagocytosis.

Fluorochrome Information:
Super Bright 436 is excited by a 405 nm violet laser and emits at 436 nm. Use a 450/50 bandpass filter or equivalent. Ensure instrument compatibility.

Buffer Recommendation:
For panels using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Target Information

SIRP alpha (CD172a) is a receptor-type transmembrane glycoprotein expressed on myeloid cells (granulocytes, DCs, macrophages, mast cells, hematopoietic stem cells). It acts as a substrate for tyrosine kinases (EGFR, PDGFR, src, insulin receptor) and negatively regulates receptor tyrosine kinase signaling. CD47 binding triggers phosphorylation of ITIMs, recruiting SHP-1 and SHP-2 phosphatases. This interaction regulates macrophage phagocytosis and T cell/DC activation. SIRP alpha influences cellular responses to serum, growth factors, insulin, oncogenes, and adhesion. Cancer cells overexpress CD47, activating SIRP alpha to inhibit macrophage-mediated tumor cell destruction.
