Product Specific Information

This 7-239 monoclonal antibody reacts with human CD169 (Siglec-1, Sialoadhesin), a type I transmembrane protein of the Siglec family.
CD169 is expressed on macrophages and dendritic cells in peripheral blood, lymph nodes, and spleen. It binds glycoproteins and glycolipids containing N-acetylneuraminyl α2-3-galactose.
Expression can be induced on CD14+ monocytes by interferon-α and interferon-γ, facilitating HIV-1 infection. CD169 also plays a role in antigen presentation to invariant NK T cells.

This antibody has been reported to inhibit erythrocyte binding to CD169-expressing cells.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and validated for flow cytometry at 5 µL (0.25 µg) per test.
A test corresponds to staining a cell sample (10⁵–10⁸ cells) in a final volume of 100 µL.

Fluorochrome Information:
Super Bright 436 is excited with the violet laser (405 nm) and emits at 436 nm.
Recommended filter: 450/50 bandpass or equivalent. Ensure instrument compatibility.

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Siglecs are membrane-bound lectins (immunoglobulin superfamily) that bind sialic acid and mediate cell-cell interactions.
Family members include sialoadhesin, CD22, and CD33.
Sialoadhesin is a macrophage-specific adhesion molecule expressed on activated macrophages in chronic inflammation and tumors.
