Product Specific Information

The eBio1D4B monoclonal antibody reacts with mouse CD107a (LAMP-1), a type I lysosomal membrane protein that is extensively glycosylated. CD107a is constitutively expressed in late endosomes-lysosomes and transiently on the surface of degranulating cytolytic T cells. It is involved in cancer metastasis and lysosomal storage disorders.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Tested on mouse thioglycolate-elicited peritoneal macrophages using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00). Recommended ≤1.0 µg/test (100 µL final volume). Cell count range: 10⁵–10⁸ cells/test. Optimize antibody concentration for best results.

Light Sensitivity: Tandem dye is sensitive to photo-induced oxidation; protect from light.
Fixation: Samples may be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-57).
Excitation / Emission / Laser: 488–561 nm / 775 nm / Blue, Green, Yellow-Green Laser.

Target Information

LAMP1 (CD107a) is a major lysosomal membrane component, with 1–2% found on the plasma membrane. It is a heavily glycosylated type I integral membrane protein involved in lysosome biogenesis and fusion with phagosomes. Upon activation, LAMP1 translocates to the plasma membrane via a tyrosine-based motif (YXXI). Surface LAMP1 promotes PBMC adhesion to vascular endothelium, suggesting a role in inflammation. Increased LAMP1 is observed near senile plaques in Alzheimer’s Disease and serves as a marker for mast cell activation.
