Product Specific Information
The eBioH4A3 monoclonal antibody reacts with human CD107a (LAMP-1), a highly glycosylated protein (~110 kDa) primarily located in lysosomal/endosomal membranes. CD107a transiently appears on the surface of degranulating cytolytic T cells and is upregulated on activated platelets and some cancer cells.
- Applications Reported: Intracellular staining followed by flow cytometric analysis.
- Applications Tested: Pre-titrated and validated for intracellular staining and flow cytometry using Jurkat cells. Recommended usage: 5 µL (0.06 µg) per test (100 µL total volume).
- Light Sensitivity: PE-Cyanine7 tandem dye is sensitive to photo-induced oxidation; protect from light.
- Fixation: Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark.
- Excitation/Emission: 488–561 nm / 775 nm (Blue, Green, Yellow-Green Laser).
- Filtration: 0.2 µm post-manufacturing filtered.
Target Information
LAMP1 (CD107a, lysosome-associated membrane protein-1) is a major lysosomal membrane component. About 1–2% is found on the plasma membrane. It contains 18 N-linked glycosylation sites, a single transmembrane domain, and a short cytosolic tail. LAMP proteins are essential for lysosome biogenesis and fusion with phagosomes.
Upon cell activation, LAMP1 translocates to the plasma membrane via a tyrosine-based motif (YXXI). Mutations in this motif affect lysosomal localization. Surface LAMP1/2 promotes PBMC adhesion to vascular endothelium, suggesting roles in inflammation. Increased LAMP1 expression is observed near senile plaques in Alzheimer’s disease and in immune cells during thymic involution. LAMP1 serves as a marker of mast cell activation.
