Product Specific Information

The DS7HAXL monoclonal antibody recognizes human Axl, a member of the TAM family of tyrosine kinase receptors (including MerTK and Tyro3). It is suitable for flow cytometry on both native and paraformaldehyde-fixed cells and can also stain formalin-fixed, paraffin-embedded tissues for IHC analysis.

• Applications Tested: Pre-diluted and tested by flow cytometry on normal human peripheral blood cells
  • Recommended use: 5 µL (0.06 µg) per test (in 100 µL final volume)
  • Applicable cell count: 10⁵–10⁸ cells/test

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vials and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days in the dark at 4°C with minimal impact on brightness and FRET efficiency.

Target Information

AXL is a receptor tyrosine kinase associated with chronic myelogenous leukemia, colon cancer, and melanoma. It transduces extracellular signals by binding growth factors such as GAS6, promoting cell proliferation. TAM receptors (Axl, MerTK, Tyro3) are also involved in phagocytosis of apoptotic cells. Deficiency of TAM receptors in mice leads to autoimmune-like phenotypes similar to systemic lupus erythematosus.

Axl may act as an entry receptor for filoviruses and is used to identify Siglec 6+/Axl+ dendritic cell subpopulations. Expression of Axl can be upregulated by TLR ligands (e.g., LPS, poly I:C). Soluble Axl is generated by proteolytic cleavage of the membrane form; increased plasma levels may indicate inflammation or cancer.
