Product Specific Information

This YCATE55.9 monoclonal antibody reacts with canine CD8 alpha. CD8α is expressed as either a heterodimer with CD8β (CD8α/β) or as a homodimer (CD8α/α). In dogs, CD8α is found on thymocytes, subsets of mature T cells, NK cells, and intestinal intraepithelial lymphocytes (IELs).
CD8 binds to MHC class I and plays a key role in thymocyte development and mature T cell activation. This antibody has been reported to inhibit cytotoxic T cell generation and function in vitro and to be immunosuppressive in vivo.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested by flow cytometric analysis of normal canine peripheral blood cells.
Use at 5 µL (0.5 µg) per test. A test is defined as the amount of antibody that stains cells in a final volume of 100 µL. Cell number typically ranges from 10⁵–10⁸ cells/test.
Excitation: 633–647 nm; Emission: 660 nm; Laser: Red Laser.
Filtration: 0.2 µm post-manufacturing filtered.


Target Information

Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin family, essential for signal transduction, T cell differentiation, and activation.
CD8 is expressed on T cells as a disulfide-linked heterodimer (CD8α/β), functioning as a co-receptor with TCR for MHC class I molecules.
CD8 is also found on subsets of T cells, NK cells, monocytes, and dendritic cells as disulfide-linked homodimers of CD8α.
Binding of MHC-I/peptide complexes by CD8 triggers Lck recruitment, leading to lymphokine production, motility, and cytotoxic T lymphocyte (CTL) activation.
Activated CTLs play a crucial role in pathogen and tumor cell clearance. Differentiation of naive CD8⁺ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
