Product Specific Information

The eBioKMC8 monoclonal antibody reacts with mouse CD9, a 24 kDa member of the transmembrane 4 superfamily characterized by four hydrophobic domains and short cytoplasmic termini. CD9 is expressed in various cell types including monocytes, macrophages, platelets, early B cells, activated B and T cells, dendritic cells, eosinophils, basophils, endothelial cells, myoblasts, and neuroblasts.

On T cells, CD9 acts as a co-stimulatory molecule. CD9 deficiency in oocytes causes sterility due to defective gamete fusion. In macrophages, CD9 associates with FcγRs to modulate phagocytosis and inflammatory signaling. In B cells, CD9 serves as a marker for marginal zone B cells, B1 cells, and plasma cells. In dendritic cells, CD9 facilitates MHC II molecule association. Expression levels may vary between donors.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse bone marrow cells; ≤0.5 µg/test recommended. One test defines the amount of antibody to stain a cell sample in 100 µL volume. Cell number should be empirically determined (10⁵–10⁸ cells/test).
Fluorochrome details: Alexa Fluor 700 emits at 723 nm, excited by red laser (633–647 nm). Recommended filters: 685 LP mirror and 710/20 band pass filter.

Target Information

CD9 antigen is a glycoprotein expressed on developing B lymphocytes, platelets, monocytes, eosinophils, basophils, stimulated T lymphocytes, neurons, and glial cells. It belongs to the tetraspanin family, spanning the membrane four times. In pre-B cells and platelets, CD9 regulates activation and aggregation, possibly through integrin CD41/CD61 (GPIIb/GPIIIa) interaction.

CD9 is involved in cell motility, osteoclastogenesis, neurite outgrowth, myotube formation, and sperm-egg fusion. It plays roles in cell attachment, proliferation, and association of MHC II molecules on dendritic cells, crucial for T cell stimulation. CD9 contributes to differentiation, adhesion, and signal transduction, and acts as a metastasis suppressor in solid tumors.
