Product Specific Information
The 53-6.7 monoclonal antibody reacts with the mouse CD8a molecule, a 32–34 kDa cell surface receptor expressed as a heterodimer (CD8αβ) or homodimer (CD8αα).
- CD8αβ: expressed on most thymocytes and a subset of mature αβ TCR T cells
- CD8αα: expressed on γδ TCR T cells, intestinal intraepithelial lymphocytes, and dendritic cells
CD8 binds to MHC class I and interacts with tyrosine kinase p56lck, contributing to T cell development and activation.
Applications Reported: Flow cytometric analysis
Applications Tested: Flow cytometry of mouse thymocyte and splenocyte suspensions (≤0.125 µg/test; 100 µL final volume). Adjust cell number empirically (10⁵–10⁸ cells/test).

Target Information
CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein receptor of the immunoglobulin family.
- Functions as a co-receptor for MHC class I molecules
- Expressed mainly on cytotoxic T cells as CD8αβ heterodimers
- Also found on subsets of T cells, NK cells, monocytes, and dendritic cells as CD8αα homodimers
- Engagement with MHC-I/peptide complexes recruits Lck kinase, inducing lymphokine production, motility, and cytotoxic T lymphocyte (CTL) activation
- CTLs are critical for pathogen and tumor cell clearance
- Differentiation of naïve CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1
