Product Specific Information
The OKT8 monoclonal antibody reacts with the human CD8a molecule, a 32–34 kDa cell surface receptor expressed as a heterodimer (CD8αβ) or homodimer (CD8αα). It is found on most thymocytes and subsets of mature T and NK cells. CD8 binds to MHC class I and associates with protein tyrosine kinase p56lck, playing a critical role in T-cell development and activation.
Preliminary testing shows OKT8, RPA-T8, and HIT8a clones do not compete for binding, indicating distinct epitopes.
This antibody is pre-titrated for flow cytometry, recommended at 5 µL (0.06 µg) per test for 10⁵–10⁸ cells in 100 µL volume.
Alexa Fluor® 700 emits at 723 nm and is excited with a 633 nm red laser (recommended filter: 710/20).

Filtered through 0.2 µm post-manufacturing filter.
Target Information
CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family, crucial for T cell activation and signal transduction.
It forms disulfide-linked heterodimers (CD8αβ) acting as co-receptors for MHC-I molecules, while CD4 serves MHC-II.
CD8 is present on cytotoxic T cells, NK cells, monocytes, and dendritic cells, facilitating antigen recognition and cytotoxic response.
Upon MHC-I/peptide binding, CD8 recruits Lck kinase, leading to cytokine production and CTL activation.
Differentiation of naïve CD8⁺ T cells into CTLs is promoted by IL-2, IL-12, and TGF-β1.
