Product Specific Information

The 2B11 monoclonal antibody reacts with mouse and human CXCR4 (CD184), also known as Fusin, LESTR, or HUMSTR. CXCR4 is a G-protein-coupled chemokine receptor that serves as a co-receptor for X4 HIV-1 entry into CD4+ cells and plays a key role in lymphocyte migration. Its ligand is SDF1.

CXCR4 is essential for maintaining granulocyte progenitors in bone marrow and for B cell development. Expression differs between species: in mice, it is restricted to CD4+CD8+ thymocytes, while in humans, expression is predominant in B lymphocytes.

The 2B11 antibody recognizes the amino terminus of CXCR4 and induces receptor internalization without blocking SDF1α binding. It is validated for flow cytometric analysis and tested on mouse thymocytes at ≤1 µg/test. Optimal staining requires ~45 minutes of incubation.

Target Information

CXCR4 (Fusin, LESTR, HUMSTR) is a seven-transmembrane G-protein-coupled receptor that functions as a co-receptor for HIV-1 and HIV-2 entry into CD4+ cells. It binds the chemokine CXCL12/SDF-1, leading to calcium signaling and MAPK activation. CXCR4 is involved in hematopoiesis, cardiac and vascular development, and neuronal survival. It also mediates LPS-induced inflammatory responses.

Antibodies against CXCR4 can inhibit HIV-1 and HIV-2 fusion and infection. The amino-terminal domain and second extracellular loop serve as key HIV binding sites.
