Product Specific Information

• Source: Monoclonal antibody obtained by fusing the P3U1 myeloma cell line with spleen cells of BALB/c mouse after immunization with soluble E-cadherin from human placenta.
• Specificity: Specifically reacts with human E-cadherin and inhibits E-cadherin–dependent cell-cell contact.
• Cross Reactivity: Does not react with rat E-cadherin.
• Storage:
  • Store at 4°C.
  • Stock solution (2.0 mg/mL) can be stored in aliquots at -20°C for 1 year.
  • After adding 0.1% sodium azide, store at 4°C for 6 months.
  • Avoid repeated freeze-thaw cycles.
• Reconstitution:
  • Dissolve lyophilized antibody in 50 µL distilled water (final concentration: 2.0 mg/mL).
  • For short-term use, dissolve directly in 500 µL or more of dilution solution.
  • Maintain antibody concentration ≥2.0 mg/mL for stability.
  • Reconstituted solution should contain 0.1% sodium azide when stored at 4°C.
• Dilution Solution Composition:
  • 20 mM TBS (pH 7.5)*
  • 10 mM CaCl₂
  • 1.0% BSA
  • (0.1% NaN₃)**
  • * Use TBS; PBS causes precipitation.
  • ** Add 0.1% sodium azide when stored at 4°C.
  • Ca²⁺ should be included to stabilize cadherin antigen.

Target Information

E-Cadherin (epithelial cadherin) is a calcium-dependent cell-cell adhesion glycoprotein belonging to the cadherin superfamily. It consists of five extracellular cadherin repeats, a transmembrane region, and a conserved cytoplasmic tail. Mutations in the CDH1 gene (chromosome 16) encoding E-cadherin are associated with gastric, breast, colorectal, thyroid, and ovarian cancers. Loss of E-cadherin function is linked to increased tumor invasiveness and metastasis. E-cadherin plays a key role in tissue architecture and integrity, and its reduction correlates with cancer progression.
