Product Specific Information

The mShad150 monoclonal antibody reacts with mouse CD150, a ~70 kDa transmembrane glycoprotein also known as Signaling Lymphocyte Activation Molecule (SLAM). CD150 is expressed by T cells (particularly Th1) and B cells, and this expression is rapidly upregulated upon activation. Immature thymocytes and dendritic cells also express this antigen.

Signaling through SLAM in T cells induces proliferation and enhances the interferon-gamma response. SLAM also plays a role in adhesion between T cells and antigen-presenting cells. The mShad150 antibody has been reported to stain the CD150⁺CD244⁻CD48⁻CD41⁻ population of pluripotent hematopoietic stem cells.

Applications Reported: Flow cytometric analysis
Applications Tested: Flow cytometric analysis of mouse splenocytes (≤0.5 µg/test). A test is defined as staining a 100 µL cell sample (10⁵–10⁸ cells/test). Antibody titration is recommended for optimal performance.

Light Sensitivity: This tandem dye is sensitive to photo-induced oxidation. Protect vials and samples from light.
Fixation: Samples can be stored in IC Fixation Buffer (Product #00-822-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information

CD150 (SLAM, Signaling Lymphocyte Activation Molecule) belongs to the immunoglobulin gene superfamily and is involved in T cell stimulation. SLAM is expressed on peripheral blood memory T cells, T cell clones, immature thymocytes, and a proportion of B cells, and is rapidly induced on naive T cells after activation.

Activated B cells express membrane-bound, soluble, and cytoplasmic isoforms of SLAM. Homophilic SLAM-SLAM binding during B–T and B–B cell interactions enhances activation and differentiation of B cells. The extracellular domain of CD150 serves as a receptor for the measles virus and acts as a co-activator on T and B cells. CD150 interacts with SH2D1A and PTPN11 via its cytoplasmic domain, and mutations in the CD150 gene may be associated with X-linked lymphoproliferative disease (XLP).
