Product Specific Information

The 14D3 monoclonal antibody reacts with human CD152, also known as cytotoxic T lymphocyte antigen-4 (CTLA-4). CTLA-4, structurally similar to CD28, is expressed on activated T cells and binds B7 family members (CD80 and CD86) with higher affinity than CD28. CTLA-4 and CD28 deliver opposing signals: CD28 acts as a costimulator, while CTLA-4 inhibits T cell activation by reducing IL-2 secretion and proliferation.

The 14D3 antibody also recognizes rhesus monkey CTLA-4 and exhibits inhibitory activity.

Applications Reported:

• Intracellular staining followed by flow cytometric analysis.

Applications Tested:

• Tested by intracellular staining and flow cytometric analysis of stimulated human peripheral blood cells using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00).

For complete detection of CTLA-4, intracellular expression should be assessed in addition to surface expression.

Light Sensitivity:

• The tandem dye is sensitive to photo-induced oxidation; protect from light.

Fixation:

• Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark.

Excitation / Emission / Laser:

• 488–561 nm excitation; 775 nm emission; compatible with Blue, Green, and Yellow-Green lasers.

Filtration:

• 0.2 µm post-manufacturing filtered.

Target Information

CTLA4 (Cytotoxic T-Lymphocyte Antigen 4) is a CD28-family receptor mainly expressed on CD4+ T cells. It binds CD80 and CD86 with higher affinity than CD28 and inhibits T cell activation. Intracellular CTLA4 is also found in regulatory T cells and plays a role in their function.
