Product Specific Information

The 9D1 monoclonal antibody reacts with mouse CD150, a ~70 kDa transmembrane glycoprotein known as Signaling Lymphocyte Activation Molecule (SLAM). CD150 is expressed by T (especially TH1) and B cells, and its expression is rapidly upregulated upon activation. Immature thymocytes and dendritic cells also express this antigen. Signaling through SLAM induces proliferation and enhances interferon-gamma response, and plays a role in T cell–APC adhesion. 9D1 is reported to be an activating antibody.

Mouse hematopoietic stem cells (HSC) can be identified using SLAM family markers (CD150⁺CD244⁻CD48⁻). For this application, antibody clone mShad150 (Product #12-1502) is recommended.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes; ≤1.0 µg/test (100 µL final volume). Optimize titration for best performance.

Light sensitivity: Tandem dye is photo-sensitive; protect vial and samples from light.
Fixation: Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-57) for up to 3 days at 4°C in the dark with minimal brightness impact.

Excitation: 488–561 nm
Emission: 775 nm
Compatible Lasers: Blue, Green, Yellow-Green

Target Information

CD150 (SLAM, Signaling Lymphocyte Activation Molecule) is part of the immunoglobulin superfamily and involved in T cell stimulation. It is constitutively expressed on peripheral memory T cells, immature thymocytes, and some B cells, and rapidly induced on naive T cells after activation. SLAM signaling enhances B cell expansion and differentiation. The extracellular domain serves as a receptor for measles virus and interacts with SH2D1A and PTPN11. Mutations in CD150 may be linked to X-linked lymphoproliferative disease (XLP).
