Target Information

The protein tyrosine phosphatase PTPN22 (PTP22, LYP, PEP, formerly PTPN8) is a genetic variant associated with risk for autoimmune diseases such as type 1 diabetes and rheumatoid arthritis.
The minor allele of a missense SNP in PTPN22 encodes a hematopoietic-specific protein tyrosine phosphatase known as Lyp.
This risk allele is found in ~17% of Caucasian individuals and ~28% of those with rheumatoid arthritis.
It may disrupt the P1 proline-rich motif crucial for interaction with the SH3 domain of CSK, potentially altering normal T cell regulation.
PTPN22 is suggested to act as an effector or regulator of CSK in T cells and other hematopoietic cells.
