Product Specific Information

This 3ZBR4S monoclonal antibody recognizes human and mouse CD247 (CD3z) when phosphorylated on Tyrosine 142. CD3 is a multi-protein complex expressed on thymocytes and T cells, serving as a marker of the T cell lineage and a critical component of T cell receptor (TCR) signaling. The CD3 zeta chain initiates the signaling cascade following TCR engagement and contains three ITAM motifs phosphorylated by Src kinases such as Lck. These phosphorylated motifs recruit SH2 domain-containing proteins like ZAP-70, amplifying downstream signaling. Loss or mutation of CD3 zeta impairs T cell development and function.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-diluted and tested on stimulated human peripheral blood cells using the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00) and standard protocols. Recommended use: 5 µL (0.5 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).
Excitation: 488 nm; Emission: 519 nm; Laser: Blue laser.

Target Information

The CD3z complex transduces antigen-recognition signals into T cells and regulates TCR complex expression. CD3 subunits (gamma, delta, epsilon, zeta) are members of the immunoglobulin superfamily encoded on human chromosome 11. Their cytoplasmic tails associate with signal transduction molecules via double tyrosine-based motifs. CD3 is present on 68–82% of normal peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells, but not on B or NK cells. Altered CD3z expression is associated with immunodeficiency disorders such as Immunodeficiency 25 and T-B+ Severe Combined Immunodeficiency.
