Product Specific Information
The 15B8 monoclonal antibody reacts with human and mouse beta-catenin, a member of the catenin family involved in cell adhesion through interaction with cadherins. Beta-catenin binds to the cytoplasmic tail of E-cadherin and participates in the canonical Wnt signaling pathway. Without Wnt stimulation, beta-catenin is phosphorylated and degraded; upon activation, it translocates to the nucleus and regulates gene expression with TCF and LEF transcription factors. Beta-catenin is expressed in thymocytes, T and B lymphocytes, and various tissues. This pathway is critical for T, B, and hematopoietic stem cell development.
Applications Reported: Suitable for intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and validated using Jurkat cells with the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00). Recommended usage: 5 µL (0.25 µg) per test in 100 µL volume. Cell number range: 10⁵–10⁸ cells/test.

Target Information
Beta-catenin is an adherens junction (AJ) protein essential for epithelial layer formation and maintenance. It interacts with E-cadherin and alpha-catenin, linking cell adhesion complexes to the actin cytoskeleton. Beta-catenin also associates with signaling complexes involving APC protein and microtubules, influencing morphogenesis and cell signaling. It is expressed in hair follicle cells and various epithelial tissues. Dysregulation of beta-catenin is implicated in cancers including colorectal, ovarian, breast, and skin tumors. In the nucleus, beta-catenin coactivates Lef/Tcf transcription factors, promoting expression of cyclin D and c-myc, which drive cell proliferation and tumorigenesis.
