Product Specific Information

The BM8 monoclonal antibody reacts with mouse F4/80 antigen (~160 kDa surface receptor) belonging to the EGF-TM7 family. It contains seven EGF-like domains, seven transmembrane segments, and a cytoplasmic domain homologous to TM7 superfamily members. F4/80 is expressed by most mature macrophages and serves as a key marker for this cell population. It may also be expressed on peritoneal eosinophils, Langerhans cells, and some dendritic cell subsets.

Expression begins early in myeloid development and can be upregulated by M-CSF stimulation. Some macrophage subsets, particularly in lymphoid tissues, may lack F4/80 expression.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on resident peritoneal macrophages; ≤1.0 µg/test recommended. Optimal titration is advised per assay.

Fluorochrome Information:
Super Bright 436 is excited by the violet laser (405 nm) and emits at 436 nm. Recommended detection filter: 450/50 bandpass or equivalent. Ensure instrument compatibility.

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific interactions.

Target Information

F4/80 antigen is a 160 kDa cell surface glycoprotein and a member of the EGF-TM7 family, sharing 68% amino acid identity with human EMR1. Expression varies during macrophage maturation and activation and is found in Kupffer cells, Langerhans cells, microglia, and macrophages in spleen, lung, thymus, and bone marrow. Some dendritic cells also express F4/80, but it is absent in T-cell zone macrophages. F4/80 may play a role in generating efferent CD8⁺ regulatory T cells.
