Description:
The 8.1.1 monoclonal antibody reacts with mouse podoplanin (T1a, gp38, aggrus), a 43 kDa transmembrane glycoprotein expressed in kidney glomerular epithelial cells (podocytes). Podoplanin is also found in epithelial and mesothelial cells such as intestinal epithelium, alveolar type I cells, and mesothelium of the visceral peritoneum. It serves as a potent marker for lymphatic endothelium and is expressed by subcapsular epithelial cells of the murine thymus. Mice deficient in Podoplanin die at birth due to respiratory defects and congenital lymphedema.

Applications Reported:
This eBio8.1.1 (8.1.1) antibody has been reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of TE-71 cell line. Recommended use ≤ 0.25 µg per test (100 µL final volume, 10⁵–10⁸ cells/test). Optimize titration for best results.

Fluorochrome Information:
Super Bright 436 is excited with the violet laser (405 nm) and emits at 436 nm. Use a 450/50 bandpass filter or equivalent. Ensure your instrument supports this fluorochrome.

Buffer Recommendation:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal fluorescence loss.

Target Information

Podoplanin (PDPN, T1A, gp38, Aggrus):
Podoplanin is a mucin-type 1 glycoprotein (40–43 kDa) expressed in tumors and normal cells, especially lymphatic epithelial cells and follicular dendritic cells. It localizes in stromal cells of peripheral lymphoid tissue and thymic epithelial cells, functioning as a regulator of lymphatic endothelium and podocyte morphology. Podoplanin acts as a ligand for CLEC-2 and is associated with tumor invasiveness and metastasis. It also plays roles in cell migration and metastasis formation. Expression is induced through Src activation of Cas and has been observed on Th17 cells. Dysfunction is linked to diseases such as lymphangioma and subglottis neoplasm. It has been proposed as a marker for lung injury.
